The technique is based on the detection of an antigen or antibodies captured on a solid surface using direct or secondary, labelled antibodies, producing a reaction whose product can be read by the spectrophotometer. The word ELISA is the acronym for “Enzyme-Linked Immunosorbent Assay”. This chapter covers the use of microplate readers for ELISA testing.
